Histological evaluation of the effects of crotoxin - Crotalus durissus terrificus in dorsal skin of Wistar rats

The crotalic venom is constituted of enzymes, toxins and peptides. Crotoxin corresponds to a greater fraction of the venom protein and is responsible for major pathophysiological changes. The main clinical manifestations of crotoxin include: neurotoxicity, myotoxicity and citotoxicity. This study aimed toevaluate histologically the local effects of crotoxin in the dorsal skin of rats. Different concentrations of crotoxin (5μg, 10μg, 20μg, 40μg and 80μg) were injected intradermally in 18 male Wistar rats. After 24 hours of injection, the skins were removed, processed and analyzed by light microscopy. Tissue samples
from the negative control group (saline) showed typical histological features of normal skin with some points of the inflammatory infiltrate and fibrin crust. All concentrations induced histological changes that have reproduced in an increasing manner and dose-dependent tissue injury. Edema and hemorrhage were heterogeous with different concentrations of the toxin. Dermonecrose was identified in the deeper regions
of the dermis, at higher concentrations. At concentrations of 10μg inflammation was characterized by leukocytosis and neutrophilia, in comparison with control. Degeneration of tissue structures was also evident in animals treated with 40μg. The collagen fibers showed little change between treatment and
control groups. However, the fibers became more eosinophilic in the groups treated with 40 and 80mg of toxin. Our results support to demonstrate the histological effects of crotoxin (PLA2) in Crotalus poisoning, which are difficult to interpret.
