In vitro germination of zygotic embryos and acclimatization of mangaba seedlings originating from the embryo culture (Hancornia speciosa)

Abstract

This study aimed to evaluate the effect of ANA and BAP on in vitro conversion of zygotic embryos of H. speciosa, and the development of efficient methods and techniques of acclimatization of mangaba derived from in vitro culture of embryos.Zygotic embryos cultured on MS medium with 0,3 g L-1 Phytagel ® and 3,0 g L-1 sucrose, combined with different concentrations of A NA (0, 0,2, 0,4 and 0,6 mg L-1) and BAP
(0, 0,5, 1 and 2 mg L-1). For the conversion of zygotic embryos of mangabeira is no need for the presence of ANA or BAP in the culture. Transplanting of seedlings was carried out in cups of 300 cm3, containing the sterilized substrates: T1- washed sand, T2- sand + dust of coconut coir (1:1) and T3-vermiculite + sand (1:1), related assessments of plant growth (seedling height, leaf number, root number and force) to ex vitro conditions. The substrates with sand and dust of coconut coir (1:1) and vermiculite and sand (1:1) resulted in greater shoot growth (2,2 and 2,8 cm), number of leaves (3,0 and 3,8) and number of nodes (1,60 and 2,2).
